pH-Sensitive Nanoparticles Developed and Optimized Using Factorial Design for Oral Delivery of Gliclazide

Gliclazide is an oral hypoglycaemic agent used for the treatment of non-insulin dependent diabetes mellitus T2DM. has low solubility in stomach and poor absorption bioavailability. The pH-dependent gliclazide influences intra- inter-subject variability. purpose this study was to develop, optimize evaluate pH-sensitive nanoparticles (NPs) based on Eudragit® S100 polymer delivery (GLZ) attempt improve its bioavailability reduce Nanoprecipitation technique preparation GLZ NPs. A 33 full factorial design applied effect independent variables (polymer concentration, volume organic phase stabilizer’s concentration) mean particle size, zeta potential incorporation efficiency developed optimal formulation evaluated using various methods including X-ray diffraction (XRD), differential scanning calorimetry (DSC), Fourier transform infrared spectroscopy (FT-IR) electron microscopy (SEM), drug dissolution glucose stimulated insulin section test. analysis results revealed transformation from crystalline unstructured form absence any chemical interactions between polymer. vitro release behaviour glucose-stimulated test showed that into NPs potentiated secretion β cells presence 10 mM glucose. Our suggests optimized have a GLZ.
